CagriSema for Type 2 Diabetes

This trial tests a new treatment called CagriSema to determine its effectiveness in lowering blood sugar in people with type 2 diabetes. Participants will receive CagriSema through one of two randomly assigned injection devices. The study lasts about 38 weeks and seeks individuals who have had type 2 diabetes for at least six months, experience frequent high blood sugar levels, and currently take metformin, a common diabetes medication. As a Phase 2 trial, this research focuses on measuring the treatment's effectiveness in an initial, smaller group of people.

You can continue taking metformin if you have been on a stable dose for at least 90 days before the study. However, you must stop any other anti-diabetic or anti-obesity medications 90 days before joining the trial.

Previous studies have shown CagriSema to be safe. Research indicates that the most common side effects involve the stomach and intestines, including mild to moderate issues like nausea or diarrhea, which usually improve over time. Notably, these studies reported no deaths, suggesting that CagriSema is generally well-tolerated by most people.¹²³⁴⁵

CagriSema is unique because it combines two active components, Cagrilintide and Semaglutide, to tackle type 2 diabetes differently from standard treatments like Metformin or Insulin. Unlike traditional therapies that primarily focus on controlling blood sugar, CagriSema aims to enhance weight loss and improve blood sugar control simultaneously. Researchers are excited about this treatment because it offers the potential for better overall metabolic health by utilizing a once-weekly injection, making it more convenient for patients and potentially leading to greater adherence and outcomes.

Research has shown that CagriSema holds promise for individuals with type 2 diabetes. In studies, it reduced HbA1c, a measure of average blood sugar levels, by 1.91 percentage points. It also significantly aided weight loss, with some patients losing up to 15.7% of their body weight. Compared to treatments like semaglutide, CagriSema achieved better blood sugar control and greater weight loss. These findings suggest it could effectively manage blood sugar and weight in diabetes. Participants in this trial will receive either CagriSema in device 1 with a placebo in device 2 or CagriSema B in device 2 with a placebo in device 1, administered subcutaneously once weekly in a dose escalation manner.¹²⁴⁶⁷
