AMG 732 for Thyroid Eye Disease
(HAZEL 401 Trial)
What You Need to Know Before You Apply
What is the purpose of this trial?
This trial tests a new treatment called AMG 732 for individuals with thyroid eye disease (TED), a condition where thyroid issues cause eye swelling and irritation. The researchers aim to determine if AMG 732 can benefit those who did not respond to previous treatments or whose symptoms returned after stopping treatment. Suitable candidates for this trial have moderate-to-severe TED and have already participated in an earlier trial with the same treatment. This trial excludes those requiring immediate eye surgery or who have undergone certain eye surgeries. As a Phase 2 trial, the research focuses on evaluating the treatment's effectiveness in an initial, smaller group of participants.
Will I have to stop taking my current medications?
The trial requires that you stop using any steroids or other non-steroid immunosuppressive agents, including monoclonal antibodies, at least 3 months before the first injection of the study drug.
Is there any evidence suggesting that AMG 732 is likely to be safe for humans?
Research has shown that AMG 732 has been tested for safety and tolerability. In earlier studies, healthy volunteers received single doses of AMG 732 under close monitoring. These studies primarily aimed to assess reactions to the treatment. Some reported side effects included injection site reactions and allergic responses, indicating the possibility of mild reactions. Overall, the treatment has been manageable. However, further research is ongoing to evaluate its effectiveness for individuals with thyroid eye disease.12345
Why do researchers think this study treatment might be promising for thyroid eye disease?
Most treatments for Thyroid Eye Disease, like corticosteroids or orbital decompression surgery, aim to reduce inflammation or relieve pressure. But AMG 732 is different because it offers a new mechanism of action by targeting specific pathways involved in the disease process. This subcutaneous injection could enhance precision in treatment, potentially reducing side effects associated with broader immune suppression. Researchers are excited about AMG 732 as it promises a more targeted approach, which could lead to more effective and safer management of the condition.
What evidence suggests that AMG 732 might be an effective treatment for thyroid eye disease?
Research suggests that AMG 732 might help treat thyroid eye disease (TED). In previous studies, researchers tested different doses of AMG 732, and the drug showed promise for TED patients. Although detailed information from these studies remains limited, this trial aims to evaluate AMG 732's effectiveness for individuals who haven't responded to other treatments or have experienced a recurrence. AMG 732 targets key pathways causing TED symptoms, potentially reducing eye bulging and swelling. More research is needed to confirm these findings, but early results are encouraging.12345

Are You a Good Fit for This Trial?
This trial is for adults with moderate-to-severe thyroid eye disease (TED) who previously took part in an AMG 732 study but did not respond or relapsed. Participants must have stable thyroid levels, not need urgent eye surgery, and agree to use contraception during and after treatment.Inclusion Criteria
