What is the purpose of this trial?

This trial tests a new treatment called ML261 (a CAR T cell therapy) for certain aggressive lung and neuroendocrine cancers. The main goal is to determine the safety of this treatment and its effectiveness in shrinking tumors. Eligible participants have previously tried other cancer treatments, experienced cancer recurrence or progression, and have a confirmed diagnosis of small cell lung cancer or specific neuroendocrine cancers. The study will also examine how the treatment moves through and affects the body. As a Phase 1 trial, this research aims to understand how the treatment works in people, offering participants the opportunity to be among the first to receive this new therapy.

Do I have to stop taking my current medications for the trial?

The trial does not specify if you need to stop taking your current medications. However, you cannot have had recent systemic anti-cancer therapies or be on certain immunosuppressive agents before starting the trial. It's best to discuss your specific medications with the trial team.

Is there any evidence suggesting that ML261 is likely to be safe for humans?

Research has shown that ML261 CAR T cell therapy, which targets a protein called DLL3, has been safe in early studies. For example, a study with mice found no major side effects. Additionally, a similar therapy targeting DLL3 caused only mild cases of an immune response known as cytokine release syndrome in a few patients, with no deaths related to side effects. Although this trial is just beginning, these findings suggest that ML261 might be safe for humans, but further research is needed to confirm this.12345

Why do researchers think this study treatment might be promising?

What evidence suggests that ML261 might be an effective treatment for small cell lung cancer?

Research shows that ML261 CAR T cell therapy targets a protein called DLL3, present in most small cell lung cancer (SCLC) tumors. Studies have found that targeting DLL3 holds promise because 85% to 94% of SCLC patients have this protein on their tumor cells. CAR T cells designed to attack DLL3 have demonstrated high effectiveness in lab tests on SCLC. Other similar treatments have shown early signs of combating tumors, with a response rate of 16.7%. These initial results suggest that ML261, the treatment under study in this trial, could help fight SCLC by specifically targeting and attacking cancer cells.14678

Are You a Good Fit for This Trial?

This trial is for adults (18+) with certain advanced or metastatic cancers, including small cell lung cancer and some neuroendocrine tumors, whose disease has come back or worsened after standard treatments. Participants must be in good physical condition and have working major organs.

Inclusion Criteria

My doctor expects me to live at least 12 more weeks.
I have a confirmed advanced or metastatic solid tumor with the required neuroendocrine features.
I am fully active or can do light work despite my illness.
