What You Need to Know Before You Apply

What is the purpose of this trial?

This trial tests a new treatment combination for a specific type of non-small cell lung cancer with the KRAS G12C genetic mutation. The goal is to determine if adding calderasib, a potential new drug, to the existing treatment of MK-3475A helps patients remain cancer-free longer after surgery. Participants will receive either the new combination or a placebo (inactive substance) along with MK-3475A. This trial may suit individuals who have had this type of lung cancer surgically removed and show no current signs of cancer based on recent scans. As a Phase 3 trial, it represents the final step before FDA approval, offering patients a chance to contribute to potentially groundbreaking treatment advancements.

Will I have to stop taking my current medications?

The trial information does not specify whether you need to stop taking your current medications. It's best to discuss this with the trial coordinators or your doctor.

Is there any evidence suggesting that this trial's treatments are likely to be safe?

Research shows that calderasib, when combined with pembrolizumab, is generally well-tolerated by patients. Previous studies have found that this combination improves outcomes for people with advanced non-small cell lung cancer (NSCLC). Although some serious side effects have been reported, they are rare. In one study, many patients experienced their cancer shrinking or stopping its growth with this treatment.

The FDA has already approved pembrolizumab for treating other cancers, indicating its safety for humans, though it can still cause side effects like any medicine. Overall, current evidence suggests that the treatments in this trial are reasonably safe for most people. However, discussing potential risks with a healthcare provider before joining a clinical trial is important.12345

Why do researchers think this study treatment might be promising for non-small cell lung cancer?

Calderasib + MK-3475A is unique because it combines two powerful approaches to tackle non-small cell lung cancer. Unlike standard treatments like chemotherapy and targeted therapies, Calderasib is a novel compound that potentially disrupts cancer cell growth by targeting specific pathways. Meanwhile, MK-3475A, also known as pembrolizumab, is an immunotherapy that helps the immune system recognize and attack cancer cells more effectively. Researchers are excited because this combination might provide a more robust and comprehensive attack on cancer cells, potentially leading to better outcomes for patients.

What evidence suggests that this trial's treatments could be effective for non-small cell lung cancer?

In this trial, participants will receive either Calderasib combined with MK-3475A or a placebo combined with MK-3475A. Studies have shown that Calderasib, when used with pembrolizumab (the active component of MK-3475A), leads to strong responses in patients with KRAS G12C-mutant non-small cell lung cancer (NSCLC). In some trials, this combination helped shrink tumors in up to 72% of patients. Calderasib targets the KRAS G12C mutation, common in lung cancer, making it a promising treatment option. Research indicates that this combination can slow cancer growth and extend patient survival. These findings suggest that Calderasib plus pembrolizumab could be effective for patients with this specific type of lung cancer.12346

Are You a Good Fit for This Trial?

This trial is for adults with a specific type of lung cancer (non-small cell, KRAS G12C mutation) that has been completely removed by surgery (stage IIA-IIIB). Participants must have no signs of cancer after surgery and be in good physical condition. Some people with well-controlled HIV or hepatitis B/C may also join.

Inclusion Criteria

Has no evidence of disease based on postsurgical radiological assessment documented by contrast-enhanced chest/abdomen CT or MRI within 28 days before randomization
Has an Eastern Cooperative Oncology Group performance status of 0 or 1 within 10 days before first dose of study intervention
Has a histological/cytological diagnosis of non-small cell lung cancer (NSCLC) with predominantly nonsquamous histology and meets one of the following criteria: newly diagnosed, treatment-naïve, resectable, clinical Stage IIA-IIIB (N2) NSCLC or completely resected, pathological Stage IIA-IIIB (N2) NSCLC including prior neoadjuvant platinum-doublet chemotherapy plus pembrolizumab or MK-3475A, or adjuvant platinum-doublet chemotherapy

Exclusion Criteria

Has diagnosis of any of the following tumor types/locations: small cell lung cancer or mixed tumors with small cell elements, neuroendocrine tumor with large cell components, sarcomatoid carcinoma, or NSCLC involving the superior sulcus
Has uncontrolled, significant cardiovascular or cerebrovascular disease within 6 months preceding study intervention
Has history of (noninfectious) pneumonitis/interstitial lung disease that required steroids or has current pneumonitis/interstitial lung disease

Trial Overview

The study compares two groups: one gets calderasib plus MK-3475A (a combination of pembrolizumab and another drug), the other gets a placebo plus MK-3475A. Both are given after surgery to see which helps prevent the cancer from coming back.
