Remibrutinib for Food Allergies
What You Need to Know Before You Apply
What is the purpose of this trial?
This trial tests a treatment called remibrutinib, a new potential drug, to determine its effectiveness for people with allergies to peanuts, cow's milk, or hen's eggs. Researchers aim to assess whether remibrutinib is safe and effective in reducing allergic reactions compared to a placebo, which is a harmless pill. Individuals who have experienced allergic reactions to these foods and tested positive for allergies might be suitable candidates. Participants will receive either the treatment or a placebo for several weeks, followed by a period where everyone receives the treatment. As a Phase 3 trial, this study represents the final step before FDA approval, offering a chance to contribute to a potentially groundbreaking treatment for food allergies.
Do I have to stop taking my current medications for the trial?
The trial information does not specify whether you need to stop taking your current medications. It's best to discuss this with the study team or your doctor.
Is there any evidence suggesting that this trial's treatments are likely to be safe?
Research has shown that remibrutinib is generally safe. In a study involving individuals with peanut allergies, most adults (87%) tolerated a significant amount of peanut protein after treatment. This suggests the drug might help reduce allergic reactions. Remibrutinib is also approved for treating chronic hives, indicating its safety in other applications. Although clinical trials always carry risks, past studies indicate that remibrutinib is usually well-tolerated, with no major safety issues reported.12345
Why do researchers think this study treatment might be promising for food allergies?
Remibrutinib is unique because it targets the Bruton’s tyrosine kinase (BTK) pathway, which plays a key role in allergic reactions. Most current treatments for food allergies, like antihistamines and epinephrine, focus on managing symptoms rather than preventing reactions. Remibrutinib offers a novel approach by potentially reducing the immune system's overreaction to allergens. Researchers are excited about this treatment because it could provide longer-lasting relief and improve the quality of life for individuals with food allergies.
What evidence suggests that this trial's treatments could be effective for food allergies?
Research has shown that remibrutinib offers promising results for treating food allergies such as peanut, cow's milk, and hen's egg. In earlier studies, it significantly reduced allergy symptoms compared to a placebo. In this trial, participants with peanut allergies will receive remibrutinib in one of the sub-studies, and it has helped many tolerate higher amounts of peanut protein. While information for cow's milk and hen's egg allergies is still being gathered, participants in separate sub-studies will receive remibrutinib to evaluate its effectiveness. Remibrutinib blocks a specific protein involved in allergic reactions, suggesting it can effectively reduce these reactions. Overall, remibrutinib is considered safe and works quickly.678910

Are You a Good Fit for This Trial?
This trial is for people aged 12 to 65 who have a diagnosed allergy to peanuts, milk, or eggs confirmed by blood and skin tests. Participants must react to small amounts of these foods and be willing to avoid them during the study.Inclusion Criteria
Timeline for a Trial Participant
Screening
Participants are screened for eligibility to participate in the trial
Treatment Period 1
Participants receive double-blind remibrutinib or placebo for up to 24 weeks in Sub-studies 1, 2, and 3, and up to 4 weeks in Sub-study 4
Treatment Period 2
Participants receive open-label remibrutinib from Week 24 to Week 52, except Sub-study 4 which starts at Week 4
Follow-up
Participants who do not enroll in the post-trial access study will enter a 4-week safety follow-up period off treatment, during which adverse events will be monitored
Open-label Extension (optional)
Eligible participants may enroll in a separate open-label post-trial access study to receive remibrutinib and evaluate long-term treatment effects

Trial Overview
The study compares remibrutinib, an experimental medication, with a placebo over one year in people with IgE-mediated peanut, milk, or egg allergies. The goal is to see if remibrutinib can safely reduce allergic reactions.
