What You Need to Know Before You Apply

What is the purpose of this trial?

This trial tests a new treatment called setidegrasib (ASP3082) for individuals with non-small cell lung cancer (NSCLC) that has a specific genetic change (KRAS G12D mutation). The goal is to determine if setidegrasib can extend the time patients live without their cancer worsening compared to the standard treatment, docetaxel. Participants will randomly receive one of these treatments. The trial seeks individuals whose cancer has spread locally or to other body parts and who have undergone up to two previous treatments. It aims to gather information on the treatment's safety, effects on tumors, and how the body processes it. As a Phase 3 trial, this study represents the final step before potential FDA approval, offering participants a chance to contribute to a treatment that could soon become widely available.

Do I have to stop taking my current medications for the trial?

The trial does not specify if you need to stop taking your current medications. However, you cannot take certain drugs that strongly affect liver enzymes (CYP3A, CYP2D6) or specific transport proteins (OATP1B1, OATP1B3). It's best to discuss your current medications with the study doctor.

Is there any evidence suggesting that this trial's treatments are likely to be safe?

Previous studies have shown that setidegrasib is generally safe for patients, meaning most people can take it without serious problems. Reports suggest that any side effects are manageable, and it shows promise in fighting cancer, especially in patients with a KRAS G12D mutation. Research has examined how the body processes setidegrasib and found it safe for people with advanced cancers.

Docetaxel is an approved treatment for several types of cancer, including lung cancer, and its safety is well understood after many years of use. Common side effects include low blood cell counts, tiredness, and hair loss, but doctors typically monitor and manage these.

Overall, human studies have shown both treatments in this trial to be safe, with setidegrasib offering a new way to treat lung cancer with specific genetic mutations.12345

Why do researchers think this study treatment might be promising for lung cancer?

Setidegrasib is unique because it targets cancer cells differently from the standard chemotherapy options like docetaxel. Unlike most treatments for lung cancer that focus on disrupting cell division, Setidegrasib specifically inhibits a key protein involved in cancer cell survival. This targeted approach could mean fewer side effects and improved effectiveness, offering new hope for patients battling lung cancer. Researchers are particularly excited about Setidegrasib because it represents a novel strategy in the fight against this disease.

What evidence suggests that this trial's treatments could be effective for NSCLC?

This trial will compare Setidegrasib with Docetaxel for treating non-small cell lung cancer (NSCLC). Research has shown that Setidegrasib may help treat NSCLC with the KRAS G12D mutation. Specifically, some studies found that 84.4% of patients had their disease controlled, and the cancer took an average of 8.3 months to worsen, indicating a delay in progression. Setidegrasib works by breaking down abnormal proteins from the faulty KRAS gene, which may help stop the cancer from spreading. Meanwhile, Docetaxel, administered to participants in another arm of this trial, is already used to treat NSCLC, but it often doesn't prevent the cancer from worsening for long.46789

Are You a Good Fit for This Trial?

This trial is for adults with advanced or metastatic non-small cell lung cancer (NSCLC) that cannot be removed by surgery and has a specific KRAS G12D gene mutation. Participants must have had no more than two prior treatments for their cancer, and cannot join if they have other targetable mutations, untreated brain tumors, or recent active cancers.

What Are the Treatments Tested in This Trial?

Interventions

  • Docetaxel
  • Setidegrasib (ASP3082)

Trial Overview

The study compares setidegrasib (ASP3082), an experimental drug targeting the KRAS G12D mutation, to docetaxel, a standard chemotherapy. Participants are randomly assigned to one of these treatments given by IV infusion. The main goal is to see which treatment helps people live longer without their cancer getting worse.
