What is the purpose of this trial?

This trial explores whether MDMA-Assisted Therapy, which combines the drug MDMA with psychotherapy, can safely and effectively improve PTSD symptoms in young adults. Participants will attend several therapy and MDMA sessions to determine if this approach alleviates their PTSD. Suitable candidates have experienced childhood trauma, meet PTSD criteria, and are willing to attend therapy sessions, abstain from certain substances, and have a supportive person to stay with them after sessions. This study aims to determine if this treatment is a viable option for people with PTSD. As a Phase 2 trial, the research focuses on measuring the treatment's effectiveness in an initial, smaller group of people.

Is there any evidence suggesting that MDMA-Assisted Therapy is likely to be safe for humans?

Research has shown that MDMA-Assisted Therapy (MDMA-AT) is generally safe and well-tolerated. This treatment combines the drug MDMA with therapy sessions and has reduced PTSD symptoms for many people. Studies reported no deaths or serious side effects related to the treatment. Some participants experienced mild side effects such as teeth grinding, anxiety, feeling jittery, headaches, and nausea, but these were not severe. Overall, evidence suggests that MDMA-AT is a safe option for treating PTSD when provided in a controlled clinical setting.12345

Why are researchers excited about this possible treatment for PTSD?

Unlike the standard treatments for PTSD, which often include antidepressants and talk therapy, MDMA-Assisted Therapy uses MDMA as an active ingredient to enhance therapeutic experiences. MDMA works differently by promoting emotional openness and reducing fear, potentially making therapy sessions more effective. Researchers are excited about this approach because it could offer faster and more profound relief from PTSD symptoms, especially for those who haven't responded well to traditional treatments.

What evidence suggests that MDMA-Assisted Therapy might be an effective treatment for PTSD?

Studies have shown that MDMA-Assisted Therapy (MDMA-AT), which participants in this trial will receive, can significantly reduce PTSD symptoms. One study found that 86.5% of participants with treatment-resistant PTSD experienced meaningful improvement in their symptoms 18 weeks after starting therapy. Another study found that combining MDMA with talk therapy improved symptoms in 67% to 71% of participants. MDMA increases certain brain chemicals, such as serotonin and oxytocin, which help regulate mood and social connections. While research is ongoing, these early results offer promise for those suffering from PTSD.12456

Are You a Good Fit for This Trial?

This trial is for young adults aged 18-26 who have PTSD related to childhood trauma. Participants must pass medical and psychiatric evaluations to ensure it’s safe for them to join.

Inclusion Criteria

I am between 18 and 26 years old.
Has someone to drive them home after each MDMA-AT session or agrees to take a taxi/rideshare while accompanied by a close other
If able to become pregnant, must have a negative pregnancy screen during study screening and on the morning of each MDMA-AT session

Exclusion Criteria

Substantial colorblindness sufficient to impede performance on fMRI tasks
Standard MRI contraindications
Has used MDMA excessively in the past

Screening

Participants are screened for eligibility to participate in the trial
