TP-05 for Lyme Disease Prevention

This trial tests TP-05, an oral medication, to determine its safety and tolerability for preventing Lyme disease, which ticks spread. The trial compares high and low doses of TP-05 with a placebo (a pill with no active drug) to assess its effects. It seeks healthy adults who frequently encounter ticks and have not recently received Lyme disease vaccines or certain treatments. Researchers are excited about TP-05 because lotilaner is a novel insecticide that could potentially prevent Lyme disease by killing ticks before they can transmit the bacteria. This oral tablet form offers a different approach by blocking the transmission of Lyme disease at its source, which could be a game-changer in prevention methods. Additionally, having both high and low dose options allows for flexibility in balancing efficacy and safety.

Research has shown that TP-05 (lotilaner) might help prevent Lyme disease. In studies with mice, TP-05 was 90% effective in preventing Lyme disease, while untreated mice showed no protection. In a human study, TP-05 outperformed a placebo by effectively killing ticks. This trial will test both high and low doses of TP-05, which in previous studies killed 89% to 91% of ticks by day 30. Earlier studies also found the treatment well tolerated, suggesting it could be a promising option for preventing Lyme disease.¹²³⁴⁵
