Felzartamab for Graves' Disease
(GRAVITATE Trial)
What You Need to Know Before You Apply
What is the purpose of this trial?
This trial tests whether felzartamab, an experimental treatment, can help people with Graves' Disease stop taking their current anti-thyroid medicine by normalizing thyroid hormone levels. Graves' Disease is an autoimmune condition where the immune system attacks the thyroid, causing it to become overactive. Participants will receive either felzartamab or a placebo through IV. Researchers will then assess if their thyroid levels remain normal without anti-thyroid drugs after 24 weeks. Individuals who have been on a stable dose of anti-thyroid drugs for at least 12 weeks and have active Graves' Disease might be suitable for this trial. As a Phase 2 trial, this research focuses on measuring the treatment's effectiveness in an initial, smaller group of people.
Will I have to stop taking my current medications?
The trial does not specify if you must stop taking your current medications, but you must be on a stable dose of anti-thyroid drugs (ATDs) for at least 12 weeks before starting. During the study, the doctor may gradually lower your ATD dose if your thyroid levels become normal.
Is there any evidence suggesting that felzartamab is likely to be safe for humans?
Previous studies have shown that felzartamab is generally safe and well-tolerated. For instance, in a study with kidney transplant patients, some experienced mild to moderate reactions during infusion, but serious side effects were rare, occurring in only one person. Another study confirmed felzartamab's safety and tolerability, although it wasn't effective for all conditions tested.
These findings suggest that felzartamab is generally well-tolerated in humans, with few serious side effects reported. However, participants should be aware that mild reactions may occur during treatment.12345Why do researchers think this study treatment might be promising?
Felzartamab is unique because it targets CD38, a protein involved in the immune response, offering a novel mechanism of action for treating Graves' Disease. Most treatments for this condition, like antithyroid drugs, focus on reducing thyroid hormone production, while felzartamab aims to modulate immune activity directly. Researchers are excited about felzartamab as it could provide a more targeted approach, potentially reducing the need for more invasive treatments like surgery or radioactive iodine therapy. Additionally, its administration via IV could ensure consistent dosing and efficacy.
What evidence suggests that felzartamab might be an effective treatment for Graves' Disease?
Research has shown that felzartamab holds promise for treating Graves' Disease (GD). Early results indicate that the drug can reduce levels of harmful antibodies attacking the thyroid gland. These antibodies cause the thyroid to overproduce hormones, leading to hyperthyroidism. Felzartamab targets specific immune cells that produce these antibodies, aiming to balance thyroid hormone levels. Previous studies have demonstrated positive results in other conditions, suggesting potential effectiveness for GD as well. The evidence supports the hope that felzartamab might help patients reduce or stop using traditional anti-thyroid drugs. Participants in this trial will receive either multiple IV doses of felzartamab or a placebo to evaluate its effectiveness in treating GD.12467

Are You a Good Fit for This Trial?
This trial is for adults aged 18-75 with Graves' Disease who have stable thyroid hormone levels on anti-thyroid drugs for at least 12 weeks, high TRAb antibody levels, and low TSH. Participants must agree not to donate blood during the study.Inclusion Criteria

Trial Overview
The study tests felzartamab (an IV drug targeting immune cells) versus placebo in people with Graves' Disease. It aims to see if felzartamab can help normalize thyroid hormones so patients can stop anti-thyroid drugs. The trial is double-blind and lasts about 9 months.
