Ontunisertib for Crohn's Disease
What You Need to Know Before You Apply
What is the purpose of this trial?
This trial tests a new treatment called ontunisertib for individuals with Crohn's disease who have bowel strictures causing symptoms like pain or cramping after meals. The goal is to evaluate how well different doses of ontunisertib ease these symptoms and reduce the need for surgery. Participants will receive either a high, medium, or low dose of ontunisertib or a placebo (an inactive look-alike treatment). This trial suits those diagnosed with Crohn's disease affecting the ileum (a part of the small intestine), who experience blockage symptoms or follow special diets, and are already on stable medication for their condition. As a Phase 2 trial, this research measures the treatment's effectiveness in an initial, smaller group, offering participants a chance to contribute to important advancements in Crohn's disease treatment.
Is there any evidence suggesting that ontunisertib is likely to be safe for humans?
Previous studies have shown that ontunisertib is generally well-tolerated by patients, with most experiencing no severe side effects. One study found that a single dose up to 1200 mg or multiple doses up to 200 mg twice daily for 10 days was safe, with no major safety issues reported. The occurrence and severity of any unwanted effects were balanced, indicating they were neither too severe nor frequent. Overall, the safety results appear promising, supporting further research.12345
Why do researchers think this study treatment might be promising for Crohn's disease?
Ontunisertib is unique because it offers a different approach to treating Crohn's Disease compared to current standard treatments like corticosteroids, immunosuppressants, and biologics. While most existing treatments aim to suppress the immune response broadly, Ontunisertib specifically targets a novel pathway, potentially leading to fewer side effects and more precise control of inflammation. Researchers are eager about its potential to provide relief for patients who may not respond adequately to traditional therapies, offering a new hope in managing this chronic condition.
What evidence suggests that ontunisertib might be an effective treatment for Crohn's disease?
A previous study showed that ontunisertib achieved promising results in treating a type of Crohn's disease that causes bowel narrowing, meeting its main goal in 103 participants after 12 weeks. Research suggests that ontunisertib could become the first treatment to prevent tissue thickening in Crohn's patients with this condition. The study also found that ontunisertib was well-tolerated, causing no major side effects. This treatment is available as an easy-to-take pill. These early findings suggest that ontunisertib might help reduce symptoms like belly pain and cramping in people with bowel narrowing due to Crohn's disease.35678
