This trial is for adults with advanced colon or rectal cancer that cannot be removed by surgery and has specific genetic changes (MSI and/or dMMR). Participants must not have received prior treatment for their advanced cancer.

Inclusion Criteria

My recent lab tests show my blood and organ function are normal.
I have tumors that can be measured on scans.
My advanced colorectal cancer is MSI-high or dMMR confirmed by testing.

Exclusion Criteria

Known Werner Syndrome (WRN)
I have an active hepatitis B, hepatitis C, or HIV infection.
I have active or uncontrolled brain cancer that needs seizure or steroid medication.

Timeline for a Trial Participant

Screening

Participants are screened for eligibility to participate in the trial

Treatment

Participants receive VVD-133214 in combination with pembrolizumab or pembrolizumab monotherapy

Regular visits for tests, scans, and check-ups

Follow-up

Participants are monitored for safety and effectiveness after treatment

Up to 36 months
30 days after last dose of VVD-133214 and/or 100 days after last dose of pembrolizumab, then every 90 days
