What is the purpose of this trial?

This trial aims to determine if Cobenfy can block the effects of cocaine in individuals with cocaine use disorder. Researchers will compare Cobenfy to a placebo to assess its safety and effectiveness. Participants will stay in a clinic for 11 days, taking either Cobenfy or a placebo, and will engage in sessions to observe how Cobenfy affects their response to cocaine. The trial seeks participants who have recently used cocaine, either by smoking or injection, and have tested positive for cocaine use. As a Phase 1, Phase 2 trial, this research focuses on understanding how Cobenfy works in people and measuring its effectiveness in an initial, smaller group.

Will I have to stop taking my current medications?

The trial requires participants to stop regular use of other medications, except for hormone-based contraceptives, daily multivitamins, short-term antibiotics, and antiretrovirals for hepatitis C. If you're taking other medications, you may need to stop them to participate.

Is there any evidence suggesting that Cobenfy is likely to be safe for humans?

Are You a Good Fit for This Trial?

This trial is for adults with cocaine addiction who are willing to stay in a hospital setting for 11 days. People with certain medical or psychiatric conditions, or those taking medications that could interfere, may not be able to join.

Inclusion Criteria

Able to speak and read English
Must meet criteria for cocaine use disorder with past month smoked or IV cocaine use and provide a urine sample positive for recent cocaine use during screening
I use birth control or meet criteria for not being able to become pregnant.

Exclusion Criteria

First degree family member with significant premature cardiac comorbidity
Chemistry values or screening outcomes outside normal ranges deemed clinically significant by study physician including lipid levels
AST, ALT or alkaline phosphatase >3x ULN
