Ketamine + Ts-134 for Healthy Subjects
What You Need to Know Before You Apply
What is the purpose of this trial?
This trial explores how different doses of ketamine, combined with a new drug called TS-134, affect brain activity in healthy individuals. Participants are divided into three groups receiving low, medium, or high doses of ketamine. Within each group, participants will take either TS-134, another treatment called XT, or a placebo. The study aims to understand the brain's response to these combinations, potentially informing future treatments for mental health issues. Individuals who are generally healthy and can commit to the study schedule might be a good fit for this trial. As a Phase 1 trial, participants will be among the first to receive this new treatment, aiding researchers in understanding its effects in people.
Do I have to stop taking my current medications for the trial?
Yes, you must stop taking any psychotropic medications (drugs affecting mood, perception, or behavior) to participate in this trial.
Is there any evidence suggesting that this trial's treatments are likely to be safe?
Research has shown that ketamine is generally safe for people, with a wide margin of safety, allowing for safe use even at higher doses. However, about 12% of people may experience "emergence reactions," such as confusion or hallucinations upon waking.
Specific safety studies for TS-134 are not yet available. This trial is in its early phase, so researchers are closely monitoring its safety for people. Early phases often focus on understanding the treatment's safety.
Overall, ketamine has been used safely for many years, but the combination with TS-134 remains under study. Participants in clinical trials help researchers learn more about any potential risks.12345Why are researchers excited about this trial's treatments?
Researchers are excited about this treatment because it pairs ketamine, a fast-acting anesthetic known for its rapid impact on mood, with TS-134 or its alternative, XT, offering a potentially new approach to managing conditions like depression. Unlike traditional antidepressants, which can take weeks to become effective, ketamine can work in just hours. The combination with TS-134 or XT might enhance or prolong these effects, offering hope for faster and possibly more sustained relief. This dual approach could address the urgent need for quicker-acting therapies in mental health.
What evidence suggests that this trial's treatments could be effective?
This trial will evaluate the effects of different doses of ketamine, both alone and combined with TS-134, on mood in healthy subjects. Studies have shown that ketamine can quickly improve mood, particularly in individuals with depression. For example, one study found that within two hours of treatment, depression symptoms decreased by about 57% to 60%, with benefits lasting at least four days. Ketamine effectively reduces symptoms even at lower doses, while higher doses provide greater benefits. Although specific data on TS-134 is limited, combining it with ketamine in this trial might enhance or prolong ketamine's effects. Overall, ketamine has shown promise for rapidly improving mood.16789

Are You a Good Fit for This Trial?
This trial is for healthy men and women ages 18-55 who do not have any major medical or psychiatric conditions, are not pregnant or breastfeeding, can use reliable birth control if needed, and can safely undergo MRI scans. People with certain health issues, metal implants, drug use history, or family history of schizophrenia cannot join.Inclusion Criteria

Trial Overview
The study tests the effects of ketamine at different doses combined with either TS-134 (or XT) or a placebo over four days in healthy volunteers. Brain activity is measured before and after treatment using special MRI scans to see how these drugs affect brain function.
