What is the purpose of this trial?

This trial tests the safety and effectiveness of a new drug, AB801, combined with chemotherapy and immunotherapy for certain types of pancreatic and bile duct cancers. AB801 helps the body's immune system recognize and attack cancer cells by blocking a protein called AXL, which promotes tumor growth and spread. The trial seeks individuals with pancreatic or bile duct cancers that have spread to nearby tissues or other parts of the body and have not been previously treated. As a Phase 1 trial, this research aims to understand how the treatment works in people, offering participants the opportunity to be among the first to receive this new therapy.

Do I need to stop my current medications to join the trial?

The trial requires that you stop taking immunosuppressive medications like corticosteroids at doses greater than 10 mg/day at least 2 weeks before starting the study treatment. Other medications that can prolong the QTc interval should be avoided if possible. For other medications, the protocol does not specify, so it's best to discuss with the trial team.

Is there any evidence suggesting that this trial's treatments are likely to be safe?

Are You a Good Fit for This Trial?

This trial is for adults with bile duct (cholangiocarcinoma) or pancreatic cancer that can possibly be removed by surgery, has spread nearby, or has spread to other parts of the body. Participants must meet specific health requirements set by the study team.

Inclusion Criteria

Creatinine clearance ≥ 50 mL/min by Modified Cockcroft-Gault formula
AST and ALT ≤ 2.5x ULN or < 5x ULN in patients with liver metastases
Hemoglobin ≥ 9 g/dL

Exclusion Criteria

Screening ECG with QTc > 450 ms
Known pregnancy, nursing women, or positive pregnancy test; WOCBP require negative pregnancy tests at screening and regularly during study
Known psychiatric or substance abuse disorders interfering with trial cooperation

Participants are screened for eligibility to participate in the trial

Treatment

Patients receive AB801 in combination with chemotherapy and immunotherapy. Cohort I cycles repeat every 21 days and Cohort II cycles repeat every 28 days for 24 weeks.

Follow-up

Participants are monitored for safety and effectiveness after treatment completion, with follow-up visits at 30 and 90 days, then every 3 months.
