What You Need to Know Before You Apply

What is the purpose of this trial?

The purpose of this research study is to determine if Metformin taken after anterior cruciate ligament reconstruction (ACLR) surgery can decrease muscle fibrosis, cellular senescence and improve leg strength. ACL injury and surgery increase muscle collagen content and inflammation which slows the recovery of muscle and function and ultimately impacts the quality of life. Individuals enrolled in the study will be randomized to receive metformin (or placebo) for 4 months after surgery. The investigators will measure muscle strength and inflammatory markers in muscle biopsy samples after the intervention period. Investigators will also follow up with participants 8 months later to determine the lasting effects of the intervention.

Are You a Good Fit for This Trial?

This trial is for people who have had ACL reconstruction surgery and are recovering from it. Participants should be experiencing muscle weakness or inflammation after their surgery. Specific inclusion and exclusion criteria were not listed.

Inclusion Criteria

BMI less than 30 kg/m2
I have a complete tear of my ACL.
My ACL surgery is scheduled within 6 months of my injury.

Exclusion Criteria

I have a bleeding disorder.
I am planning to have ACL surgery using a quadriceps tendon graft.
I have an infection near my lower leg or foot.

Interventions

  • Metformin

Trial Overview

The study tests whether taking metformin for 4 months after ACL surgery helps reduce muscle scarring, aging of cells, and improves leg strength compared to a placebo. Participants are randomly assigned to either the metformin or placebo group.
